Ethanol extract of African Leaf effect on MDA and SOD level in rats induced Doxorubicin

Abstract

The African leaves or Vernonia Amygdalina is a popular herb plant used as a traditional medicine in Asia and Africa. African leaves apparently have many benefits for health that many people have not known, include lowering cholesterol, help cancer treatment, preventing heart disease, relieves malaria fever symptoms, contains antioxidants, helps diabetic treatment. This leaf extract contains active substances such as flavonoids, alkaloids, polyphenols, saponins, and tannins. Doxorubicin (Dox) is anticancer agent, but unfortunately also have damage effect as free radical. The goals of this experiment is to identify the extent to which ethanol extract of African leaf (EEAL) affects the increase in superoxide dismutase (SOD), an enzyme that helps break down potentially harmful oxygen molecules in cells, and how much decreased malondialdehyde (MDA) levels in Doxorubicin - induced rats, MDA is a reactive compound that is formed naturally and is a marker of oxidative stress.Twenty four male wistar rats were divided into 6 treatment groups. Group 1 Normal, 2 Dox-induced, 3 Dox + vitamin C, 4. Dox + EEAL 100 mg/kgBW, 5 Dox + EEAL 300 mg/kgBW and 6 Dox EEAL 500 mg/kgBW. The result was SOD activity increase about 3-6 mg/ml after administration of ethanol extract of African leaves, compared to the group Dox induced without EEAL. The level of MDA reduce about 0,3 - 2 mg/mL after administration of EEAL 100 mg, 300 mg and 500 mg./kgBW.
