Abstract

Objective: Comparison Study of Pain Relief Efficacy, Inflammation, and Healing between Metamizole Sodium and Meloxicam on Deep Dermal Burns of Rats Wistar Using IL-6 Reduction and Histopathology Wound Healing. Methods: The design of experiment used post-test control group, which consisted of 18 adult male Wistar rats, aged 10 w, weighing 150-250 gs, namely three groups such as metamizole sodium group, meloxicam group, and control group using aquadest (n=6 in each group). The method of deep dermal burn induction was carried out by pressing the aluminum rod heated to 100 degrees Celsius for 15 seconds. Both drugs were administered through oral route based on the body weight of rats (360 mg/kg/day for metamizole sodium; 0.675 mg/kg/day for meloxicam). On day 3, blood samples were measured for the IL-6 level by ELISA method and histopathological study of wound biopsy through HandE and Masson’s Trichrome Staining. Results: The initial concentration of IL-6 was comparable across the groups being 107-108 pg/ml. Following the three-day treatment, the levels of IL-6 were 89.9 pg/ml (metamizole), 87.5 pg/ml (meloxicam), and 95.7 pg/ml (control). In terms of percentage decrease of IL-6 from the initial levels, metamizole, meloxicam, and control had 16.9%, 19.0%, and 11.6% reduction, respectively. According to ANOVA results, there was no statistically significant difference across the groups(F=2.547, p=0.112). As illustrated by histopathological scores, there was a better epithelialization, reduced inflammatory infiltrates, and improved collagen deposition in the case of meloxicam (9.17) compared to metamizole (8.5), and control (8.83). Conclusion: Both drugs have demonstrated effective pain relief and decrease of IL-6 levels, but meloxicam proved superior in terms of histopathological scores.
