This chapter explores the key challenges in biologics drug delivery, including their low permeability across biological membranes and their immunogenic nature. Various routes of administration are discussed, each offering distinct advantages and limitations. The central focus of this chapter is the application of polymers in enhancing the delivery of biologics. Both natural and synthetic polymers are evaluated for their applications in delivering biologics. Additionally, a variety of synthetic polymers, including poly(ethylene glycol) (PEG), poly(lacticco-glycolic acid) (PLGA), polylactic acid (PLA), and dendrimers, are explored for their role in protecting biologics from degradation, enhancing permeation, and controlling release kinetics.
